Katherine Parkinson estudió literatura clásica en la Universidad de Oxford antes de formarse en la Academia londinense de música y arte dramático.

En la pequeña pantalla, se ganó al público británico con su papel de coprotagonista, Jen, de la comedia de Channel 4 The IT Crowd, y dando vida a Pauline en la serie de ITV Doc Martin. Ha sido también estrella invitada en Ahead of the Class (interpretada por Julie Walters), Christmas at the Riviera, y personaje habitual en Katy Brand’s Big Ass Show de ITV2, dado que es amiga de Brand desde sus tiempos estudiantiles en Oxford.

En cuanto a su trayectoria teatral, ha dado vida a Masha en La gaviota en el Royal Court Theatre con Kristin Scott Thomas, The Lightning Play en el Almeida Theatre, Other Hands en el Soho Theatre, The Unthinkable en el Sheffield Crucible Studio, Cigarettes & Chocolate en el Kings Head Theatre, Flush en el Soho Theatre, The Riot Act y The Increased Difficulty of Concentration en el Gate Theatre, y The Age of Consent en el Festival de Edimburgo y el Bush Theatre, entre otros.

Tras completar Easy Virtue, Katherine rodó la película de Richard Curtis The Boat that Rocked, con Philip Seymour Hoffman, Bill Nighy y Tom Sturridge.
